Application and Introduction to Liquid Capsule Filling Technology and Equipment---I

Traditional capsules can be divided into two types: hard capsules (be filled with powdery medicine) and soft gels (be filled with liquid medicine such as fish oil, Vitamin E and Vitamin A). In recent years, new liquid capsule filling technologies have been developed, such as filling semi-solid or oily liquid in hard capsules. Among them, full-transparent liquid capsule filling technology is completely new. However, it provides a simple but effective method for medicine and healthcare products development although it is still in progress.

 
1. Introduction and development for liquid capsule
The development of capsules has a history of thousands of years. 1500 B.C. saw the advent of the first capsule in Egypt. Since then the research on capsules had been progressing slowly. And the start of industrial production made development of capsules dramatically rapid.
 
In 1730, a Vienna pharmacist produced capsules with starch;
In 1834, capsule filling technology was patented in Paris;
In 1846, two-section-type capsule filling technology was patented in France;
In 1872, the first capsule filling machine was manufactured in France;
In 1874, industrial production of hard capsule started in Detroit and variable capsule sizes were introduced;
In 1888, hard capsule filling technology of Parke-Davis is patented in Detroit;
In 1931, the capsule production speed of Parke-Davis reached 10,000pcs/h;
In 1935, soft gel was invented by General Nutrition Centers (GNC);
In 1984, the first capsule sealing machine was invented by Shonogi, a Japanese enterprise which was instructed by Eli Lilly and Company.
 
When it stepped into the 21st century, liquid capsule, as a new form of healthcare product, was developed in Japan and USA, followed by wide application of sealing technologies. Currently, an obvious trade is that OTC and healthcare product enterprises drive sales growth with new drug form development. Apparently, OTC is increasingly becoming a critical part of global medical market. Considering high investment and crisis of API development, there is no doubt for new drug form development with low investment crisis and short return time to be strong power to drive expansion of OTC and healthcare production line.
Liquid capsules, or liquid-filling hard capsules are two-section. They are specially designed to strengthen seal and maintain liquid volume. The capsule shells are made from gelatin or HPMC and hailed as the ideal container for liquid and semi-solid preparations. By this way can the liquid medicine be released quickly so that nutrient can be delivered to the body in time.
 
2.Background of Liquid-Filling Capsule Development
Application of new drug form can make the product more competitive. Innovation of OTC and healthcare products can be advanced by high-efficiency and convenient preparation forms. How to improve absorption of insoluble drugs is a key problem for today’s medial technologies to tackle. It is necessary to develop liquid-filling hard capsules since they are compatible with various excipients and can improve the solubility and bioavailability of the nutrients. Liquid-filling capsules are of low oxygen transmission and are degradable. Besides, they can have wide application in other fields.
 
With the global healthcare product boom, a large number of natural liquid healthcare products sell well on the market. Correspondingly, transparent liquid-filling capsules draw attention of more and more healthcare product manufacturers. Soft gels are made from traditional gelatin and thus are not completely transparent and hardly represents the features of green healthcare products. However, foreign companies invented transparent liquid-filling hard capsules. They are made from material excluding gelatin and have high transparency, drug resistance, moisture resistance and durability.
