What is pharmaceutical pellet medicine

When drugs and blockers are mixed, processed to pills and then coated, we can get oral solids. One kind of oral solids is pellet. It can also be divided into two types, namely sustained-release pellets and control-release pellets. Pellets’ diameter is usually less than 2.5mm. Normally, sustained-release pellets can be filled in capsules to produce sustained-release pellet capsules.
 
Pellet production technologies have had a rapid development since 1950s. In recent years, development of a variety of excipients also have enabled sustained-release pellet to show its exclusive advantages and to be known as one of the ideal sustained-release dosage form. Many pharmaceutical enterprises developed such sustained-release pellets, e.g., Contac sustained-release capsules produced by SK&F, Aspirin sustained-release capsules produced by CPU-Pharma, and Olfen-100 which was improved for import to China in 1997.
 
Pellet is dispersive dosage form. Its single dosage is composed of multiple units. Therefore, it has some advantages compared with other single-dosage forms.Please check the advantage of medicine pellet as following:

 
1) More contact area of drugs with gastrointestinal tract, absolute absorption and higher bioavailability.
2) Ideal drugs releasing rate by combination of pellets with different rate, expected drugs concentration in blood, avoidance to peak and valley phenomenon, longer action time and less side effects.
3) Uniform distribution of drugs in pellets and lower irritation to gastrointestinal mucosa.
4) Defects of some pellets will not have serious influences on the whole drugs releasing action and will not cause dosage failure as tablets since the releasing ability is the sum of multiple pellets in a unit dosage and drug dose is dispersed in all of these pellets.
5) Pellets can distributed in the whole gastrointestinal tract, be less influenced by changes of gastric emptying function (such as pylorus closure), overcome the time difference of transfer in gastrointestinal tract and effect difference caused by irregular gastric emptying, avoid problems of tablets, i.e., irritation caused by their unexpected retention.
6) Different drugs can be processed to pellets and these different pellets can then be compounded to preparations. Such preparation are featured by stable effect and convenient quality control.
7) Some drugs’ properties can be changed when they are processed to pellets. For example, pellets are of good fluidity, uniform size, easy processing (coating, tablet pressing or capsule filling).
8) By processing to pellets, some drugs’ stability are improved and uncomfortable smells are covered.
9) It’s easy to process pellets to sustained-release, control-release or targeting preparations.
 
All in all, development of pellet medicine is becoming a branch of sustained-release or control-release preparation study and research.
